摘要:
目的 研究苍苓散对轮状病毒(rotavirus, RV)肠炎患儿肠道菌群的影响。方法 收集9 例RV 肠炎患儿在 治疗前及治疗5 d 后的粪便,采用16S rDNA 测序法进行生物信息分析。结果 苍苓散治疗前后的肠道菌群优势物种极其 相似,但所占比例不同;2 者在门水平上均以厚壁菌门的丰度为最高,治疗前脱硫弧菌目、脱硫弧菌科、瘤胃菌科、嗜 胆菌属、隐秘杆菌属丰度较高,治疗后乳杆菌目、乳杆菌科、乳杆菌属丰度较高,其差异有统计学意义(P < 0.05)。 用香农指数、辛普森指数所估算的微生物群落多样性,UniFrac 聚类分析判断的物种组成相似性,以及PCoA、Anosim 分析及物种多响应置换过程分析等,均未能发现治疗前后的肠道菌群结构存在显著差异。结论 苍苓散的治疗,影响了 RV 肠炎患儿肠道中某些菌群的丰度,但需要更大样本、更深入的对照研究以明确中医药治疗婴幼儿RV 肠炎影响肠道菌 群的具体机制。    
Abstract:
Objective To explore the effect of Cangling Powder on intestinal flora in children with rotavirus (RV) enteritis. Methods The feces of 9 children with RV enteritis before and 5 days after treatment were collected for bioinformatics analysis by 16S rDNA sequencing. Results The dominant species of intestinal flora before and after treatment of Cangling Powder were very similar, but the proportion was different. Both of them had the highest abundances at the p_Firmicutes, and the abundances of o_Desulfovibrionales, f_Desulfovibrionaceae, f_Ruminococcaceae, g_Bilophila and g_Faecalibacterium were higher before treatment. The abundances of o_Lactobacillales, f_Lactobacillaceae and g_Lactobacillus were higher after treatment, and the difference was statistically significant (P < 0.05). The diversity of microbial community estimated by Shannon Index and Simpson Index. The similarity of species composition by UniFrac cluster analysis, and PCoA, Anosim analysis and species multi-response permutation procedures analysis showed no significant differences in the intestinal flora structure before and after treatment. Conclusions Cangling Powder treatment affects the abundance of some bacteria in the intestinal tract of children with RV enteritis. Larger sample size and indepth comparative studies are needed to clarify the intestinal microbiota mechanism of traditional Chinese medicine treatment in children with RV enteritis.
