摘要:
目的 探讨人肠道病毒71 型(enterovirus 71, EV71)结构蛋白VP1 和RNA 依赖性RNA 聚合酶(RNA-dependent RNA polymerase, RdRp)诱导健康成人外周血T 细胞产生IFN-γ 的应答情况。方法 通过重叠肽技术合成覆盖EV71 VP1 和RdRp 蛋白的重叠肽段作为抗原,采用IFN-γ 酶联免疫斑点分析法和磁珠分选法体外检测40 例健康成人VP1 和RdRp 抗 原特异性CD4+ 和CD8+ T 细胞免疫应答特征。结果 40 例健康成人中有12 例(30%)检测到RdRp 蛋白特异性T 细胞反 应,RdRp 蛋白诱导的免疫应答完全以特异性CD4+ T 细胞为主;而未检测到VP1 特异性T 细胞免疫反应;同时筛选出9 个 RdRp 蛋白特异性CD4+ T 细胞表位。结论 健康成人对RdRp 蛋白存在特异性T 细胞反应,且以特异性CD4+ T 细胞为主, 该研究有助于阐明EV71 感染的免疫学应答机制,并为EV71 感染防控提供指导价值。 
Abstract:
Objective To explore the responses of IFN-γ induced by enterovirus 71 (EV71) capsid protein VP1 and RNAdependent RNA polymerase (RdRp) in peripheral T cells of healthy adults. Methods The characteristics of VP1 and RdRp specific CD4+/CD8+ T cell immune responses in 40 healthy adults were analyzed by using IFN-γ enzyme-linked immunospot and magnetic cell sorting assays, with overlapping synthetic peptides covering EV71 VP1 and RdRp protein as the antigen. Results Among 40 healthy adults, 12 cases (30%) showed EV71 RdRp specific T cell responses, mainly CD4+ T cells. No T cell responses against VP1 were detected in healthy adults. In addition, 9 individual peptides containing potential RdRp specific CD4+ T cell epitope regions were identified. Conclusions T cell response specific for RdRp protein is observed in healthy adults, mainly CD4+ T cells. This study is conductive to elucidate the immunological response mechanism of EV71 infection and provides guidance for EV71 infection prevention and control.
